Optimal caching system?

One of the projects is growing and it's time to add an additional layer of caching.

Now available:

Load balancer -> N front-ends -> memcached -> MySQL


Planned:

Load balancer -> WEB cache -> N front-ends -> memcached -> MySQL


As a WEB-cache can be Squid, Varnish, nginx.


Are there any subtleties and pitfalls in this architecture?

What would you advise for optimization.

For reliability, better Load balancer -> M WEB-caches -> N front-ends -> memcached -> MySQL

I agree, but I'm interested in in the first place, the possible pitfalls in this implementation. Excessive caching can also be damaging.
It would be interesting to hear the opinion of those who implemented it in practice.
